Effects of fibroblast growth factor (FGF acidic) and epidermal growth factor (EGF) on procollagen-N proteinases; ADAMTS-2 and ADAMTS-3 in osteosarcoma model

A Disintegrin and Metalloproteinase domain, with Thrombo-Spondin (ADAMTS) is a family of zinc-dependent proteases thathave common features with A Disintegrin and Metalloproteinase(ADAM) and also MMP enzymes (Matrix Metalloproteinases).ADAMTS’s have important roles in a variety of normal condi-tions such as development, homeostasis, fertility and pathologicalconditions, including arthritis and cancer.ADAMTS-2 and ADAMTS-3 cleave type I, II, III and V col-lagen precursors into mature molecules. ADAMTS-2 has impor-tant functions both in regulating the formation and structure ofskin and in the maturation of spermatogonia. It is correlate withthe incidence of both Ehlers–Danlos syndrome type VII C anddermatosparaxis in cattle. ADAMTS-3 has been identified as atype II procollagen N-propeptidase, It’s expression is much lowerthan ADAMTS-2 in skin but is about 5-fold that of ADAMTS-2in cartilage.In this study we examined the effect of Epidermal GrowthFactor (EGF) and Fibroblast Growth Factor (FGF acidic) onthe mRNA expression level of ADAMTS-2 and ADAMTS-3 in aosteosarcoma model, SaOs-2 cells. This effect has been invesiti-gated by quantitative RT-PCR (qRT-PCR) in diffferent timeintervals namely 1, 2, 3, 6, 24, 48 and 72 hour. Acidic FGF upre-gulates ADAMTS-2 and ADAMTS-3 expression at mRNA levelsup to four fold in 6 hours. EGF upregulates ADAMTS-2 andADAMTS-3 up to two fold for 24 hours.
